Understanding the effect of digestion on oral, lipid-based drug formulations using Rainbow and MicroDISS

A new paper entitled, “Developing an in vitro lipolysis model for real-time analysis of drug concentrations during digestion of lipid-based formulations,” was recently published in the European Journal of Pharmaceutical Sciences. The paper discusses the importance of understanding the effect of digestion on oral, lipid-based drug formulations in the intestine.

The paper, concludes, in part:

“This study demonstrated the viability of using…the MicroDISS ProfilerTM..

While the main application of this method is seen in the context of high throughput screening and not as a direct alternative to the classical pH-stat method, the application presented herein demonstrates that a direct estimate of the drug solubilization capacity of lipid dispersion and digestion media can be obtained using in-line measurement.

In contrast, in order to generate equivalent estimates of the solubilization of lipid dispersion and/or digestion using the classical setup, this would entail a series of blank digestion experiments, sample extraction, addition of lipolysis inhibitors, addition of excess drug and followed by solubility studies over time to determine comparable solubilization capacity.

The benefit of the MicroDISS ProfilerTM is readily apparent in terms of higher throughput and smaller sample size.”
